e-tender notice inviting tender for small rna sequencing of wheat samples as per tender document , contract for small rna sequencing of wheat samples , specifications for small rna sequencing of wheat samples1 . a total of 24 samples of wheat tissuesto be processed 2 . quality control of samples should be done using qubit& agilent bioanalyzer , rin ( rna integrity number ) and values should be at least 7-8 . 3 . required mrna library preparation of wheat samples using illumina platform with 1x75 single end sequencing chemistry . 4 . a total of 20-25 million reads ( or more ) of >q30 . need to generate 1-2gb data per sample . 5 . carry out bioinformatics analysis for these samples which includes:a ) quality checking of sequence reads , filtering of high-quality sequence data and assembly of contigs . following quality checks will be performed ( i ) base quality score distribution , ( ii ) sequence quality score distribution , ( iii ) average base content per read , ( iv ) read length distribution , ( v ) biasing of k-mers . b ) small rna assembly and annotation ( mirna , sirna , snorna , pirnaetc ) , target prediction . raw data with quality and statistical analysis . c ) reporting of known small rna ( mirna and other non coding rnas ) . prediction of novel small rnas . prediction of mirna/small rna targets , mirna based ssr markers alongwith primers list , tasi-mirnas etc . d ) mirna network/hub genes for salt stress , mirna target plot and heat map of differentially expressed genes . e ) if data does not fulfil the above criterion , data will not be accepted . sample details: 24 samples will be sent to the firm in split ( i . e . 12 samples in current financial year and 12 samples in the next year . it will be the responsibility of the service provider to collect the samples from the lab and transport to the sequencing centre under proper conditions at its own cost . the institute will not bear any cost for trasport of samples .
