Global Tender Document For Colony Picking Robot; System Capabilities and Technical Specifications Imaging and Detection Capabilities • Must automatically detect colonies of algal, yeast, filamentous fungi, and bacterial organisms. • Must include a minimum of 6 lighting channels, enabling: o Fluorescence imaging across visible and UV spectrum o Detection of fluorophores including GFP, mCherry, and BFP Picking and Pinning Capabilities • Must support picking from and pinning to both liquid and solid agar plates. • Picking speed: 600 colonies per hour or more. • ZOI (Zone of Inhibition) measurement capability should be available. • Must use a non-metallic disposable picking surface: o Prevents cross-contamination o No requirement for washing or sterilisation procedures • Capable of picking various colony types without head or pin changes o Must achieve >95% transfer rates with environmental samples/isolates • Accuracy of picking must be to 50 μm. • Auto-offset functionality for multiple picks from the same colony. • Ultrasonic agar surface detection and contact pressure regulation: o Enables consistent picking on uneven agar surfaces Precision, Compatibility, and Formats • <9 g/mm² surface load applied during picking • Must support: o 96, 384, 1536, and 6144 format array densities • Must include streaking functionality: o Customisable streak length and shape • Must include wet mix functions: o Re-suspension and homogeneous transfer of cells from liquid cultures • Must support: o SBS-format plates (e.g., Nunc Omnitray, 6–96 well, deep well, PCR plates) o Petri dishes (9 cm and 15 cm) o MALDI-TOF slides Data Management and User Interface • Suitable Computer systems to be provided • User interface must be intuitive, with 90% of functions learnable in <10 minutes. • Individual user login and password to support user credential and data security • Must allow filtering based on: o Colony size, roundness, proximity, brightness, contrast, RGB colour, Cartesian/Cardinal coordinates • Must support: o Fully customisable plate sectoring templates o Special mode for CSV-based custom protocols (e.g., Excel) o Complete data traceability: CSV exports including colony parameters, coordinates, pre- and post- processing images API and Integration Requirements • Must include a free, supported API (no extra cost) • Must support integration with robotic arms • At least one instrument must include or be upgradable to include an automatic door compatible with robotic arms Performance Guarantees • Transfer efficiency: o 99.5% for E. coli and yeast o 95% for filamentous fungi and algae • Cross-contamination risk < 0.05% • No ethanol-based cleaning system required Support and Servicing • Technical support must: o Respond to ≥85% of queries within 24 hours (weekdays) o Respond to ≥43% of queries within 1 hour • Technicians must be trained and certified to support: o Colony picking systems o Colony imaging systems
