Degenerative Aortic Valve Stenosis (As) Is The Most Common Fatal Heart Valve Disease In Industrialized Countries And Affects Over 1 Million Women And Men In Germany Alone With A 5-Year Mortality Rate Of Up To 80%. To Date, There Is Not A Single Drug Available Worldwide For The Treatment Or Prevention Of As. The Only Treatment Currently Available Is Replacing The Diseased Valve With A Prosthesis. This Is Not Only Stressful For Patients, But Also Involves High Costs. These Currently Amount To Over 2 Billion Euros Annually, In Germany Alone. In The Past, Some Attempts Have Been Made To Develop Drugs For The Treatment Or Prevention Of As. All Of These Attempts Have Been Unsuccessful To Date. Through Our Fundamental Work In The Dfg-Funded Collaborative Research Center Aortic Diseases (Trr 259), We Were Able To Show That Various Mechanisms That Lead To The Development Of As Can Be Inhibited By Activating The Receptor Liver X Receptor (Lxr). Allstars Develops Well-Tolerated, Selective And Highly Potent Lxr Ligands.
