The Desired Service The Maternally Expressed Gene 3 (Meg3) Encodes An Lncrna (Long Non-Coding Rna) With A Tumor Suppressor Effect Which Modulates The Responses Of The P53 Protein. The Structure Of This Lncrna Contains Two Evolutionarily Conserved Distal Motifs That Interact Through Base Complementarity To Form Mutually Exclusive Pseudoknot Structures (See Figure). Mutations That Disrupt These Interactions Have Been Shown To Impair Meg3-Dependent Stimulation Of P53 In Vivo And Disrupt Meg3 Folding In Vitro [1]. The Required Service Consists Of The Production Of Synthetic Lncrna Sequences In Biologically Active Conformations That Recapitulate Meg3 Pseudoknot Structures In A Chemical Form Allowing Their Encapsulation Into Controllable Liposomal Carriers For Delivery Into Cancer Cell Organoids And Murine Xenografts. Mutant Forms (Disrupted Interactions) Will Also Be Produced For Use As A Negative Control. The Production Protocol Will Integrate Non-Denaturing Purification Of Lncrnas With In-Solution Hydrodynamic Analysis And Atomic Force Microscopy (Afm) Imaging To Produce Highly Homogeneous Lncrna Preparations, Monitored By Visualization Of Their 3D Topology At ~15 Å Resolution [2]
